HIV Drug Reduces Biological Age in Healthy Adults by Targeting Retrotransposons

Researchers tested whether an FDA-approved antiretroviral drug could slow biological aging in healthy, HIV-negative adults. The study focused on tenofovir alafenamide (TAF), which blocks reverse transcriptase—an enzyme used not only by HIV but also by retrotransposons, mobile genetic elements that become more active with age and may drive inflammation and cellular damage. In this small proof-of-concept trial, participants taking TAF showed measurable reductions in biological age markers compared to controls. This is the first human evidence that targeting retrotransposons might be a viable anti-aging strategy. While the sample size is modest and this is a preprint (not yet peer-reviewed), the findings are significant because they use an already-approved, well-characterized drug and suggest a entirely new mechanism for gerotherapeutics. If confirmed in larger, longer studies, retrotransposon inhibition could join senolytics and other interventions as a practical approach to slow aging. The safety profile of TAF is well-established from HIV treatment, which could accelerate clinical development for longevity applications.
