A fascinating new study reports that the collapse of nocturnal melatonin amplitude—the nighttime surge in this key hormone—correlates with biomarkers of both microbiome disruption and glymphatic system dysfunction, and critically, this relationship holds independent of chronological age. The glymphatic system is the brain's waste-clearance pathway, crucial for removing toxic proteins like amyloid-beta. The findings suggest that melatonin decline may be a central node connecting multiple hallmarks of biological aging: circadian disruption, gut dysbiosis, and impaired brain clearance. This matters because melatonin amplitude loss occurs at different rates in different people, potentially explaining why some individuals age faster than others. If melatonin collapse is causally involved—not just correlated—interventions targeting circadian rhythm restoration or melatonin supplementation might simultaneously improve gut health and brain clearance.
