Mushroom Compound Inotodiol Prevents Age-Related Muscle Loss via Mitochondrial Rescue
Researchers have identified inotodiol, a natural compound from the medicinal mushroom Inonotus obliquus (Chaga), as a promising intervention against age-related muscle wasting (sarcopenia). Published in Signal Transduction and Targeted Therapy, the study demonstrates that inotodiol prevents muscle loss by restoring mitochondrial function through activation of liver X receptor beta (LXRβ) signaling. Sarcopenia affects up to 15% of people over 65 and substantially increases mortality risk, but therapeutic options remain limited. The researchers showed that inotodiol treatment improved mitochondrial respiration, reduced oxidative stress, and maintained muscle mass and strength in aging animal models. The LXRβ pathway represents a novel target for sarcopenia, distinct from the more commonly studied mTOR or AMPK pathways. While these are preclinical findings requiring human validation, the results are compelling because mitochondrial dysfunction is a hallmark of both aging and sarcopenia. Chaga mushrooms have been used traditionally for various health purposes, but this work provides a specific molecular mechanism. If confirmed in human trials, inotodiol could become a natural supplement or drug candidate for preserving muscle function during aging.
