NMN Supplement Boosts REM Sleep by Activating Brain Rhythm Neurons

A new mechanistic study reveals that nicotinamide mononucleotide (NMN), a popular longevity supplement and NAD+ precursor, directly stimulates the activity of bursting slow-oscillation neurons in the supramammillary nucleus of the brainstem, leading to enhanced REM sleep. REM (rapid eye movement) sleep is crucial for memory consolidation, emotional regulation, and brain waste clearance. The study identifies a specific cellular pathway through which NMN affects sleep architecture, moving beyond general metabolic effects to show targeted neuronal activation. This matters because sleep quality, particularly REM sleep, declines with age and is strongly linked to cognitive health and dementia risk. While many NAD+ boosters are taken for metabolic benefits, this research suggests NMN may also improve brain function through sleep enhancement. The findings are particularly relevant given growing evidence that sleep disturbances accelerate brain aging. However, this appears to be a preclinical study, so human trials specifically measuring sleep architecture with NMN supplementation will be needed to confirm these benefits translate to people and at what doses.
