KDM4C Inhibition Boosts NK Cell Attack on Leukemia via Immune Pathway Activation

Researchers have found that inhibiting KDM4C, a histone-modifying enzyme, significantly strengthens natural killer (NK) cells' ability to destroy acute myeloid leukemia (AML) cells carrying TP53 mutations. The mechanism works through the cGAS-STING pathway, a key immune sensing system that detects cellular stress and triggers anti-tumor responses. TP53 mutations are notoriously difficult to target and associated with poor outcomes in AML, so finding ways to enhance the body's own immune cells against these cancers is particularly valuable. While this study appears focused on cancer treatment rather than aging per se, the cGAS-STING pathway is increasingly recognized as central to both immune aging and cellular senescence. Understanding how to modulate this pathway—whether to enhance anti-tumor immunity or potentially manage chronic inflammation in aging—represents an important area of convergence between cancer and longevity research. The findings may eventually inform strategies to reinvigorate immune function in older adults, though clinical translation remains distant.
